Inhibition of Ovarian Carcinoma Growth in Nude Mice by Administration of Gonadotropic Hormone-Releasing Hormone Analog

Abstract
Human ovarian epithelial carcinoma cell lines were heterotransplanted in female nude mice and growth was evaluated in intact and gonadotropin-releasing hormone (GnRH) analogue [D-Trp^6]treated mice. We observed following valuable results. 1. Chronic administration of GnRH analogue to mice bearing measurable ovarian carcinoma significantly decreased tumor volume compared to control group. 2.Early administration of GnRH analogue to mice after injection of tumor cells showed significant early inhibitory effect on tumor formations. 3. Administration of GnRH analogue to mice after intraperitoneal injection of tumor cells showed inhibitory effect on ascites and multiple visceral tumor formation. 4. Mice serum CA-125 level showed decreasing tendency in GnRH analogue-treated mice group, but statistically non-significant. 5. Morphological difference including remarkably reduced in cellularity and increased in fribroblastic stroma was found on tumors developed in GnRH analogue-treated group. 6. Administration of GnRH analogue to mice increased the natrual killer cell activity compared to control group. Regression of ovarian tumors in nude mice in response to chronic administration of [D-Trp^6] suggests that this compound should be considered for the new therapeutic possibilities toward the control of neoplastic ovarian growth in women.
